Heat Denaturation of DNA

   

To produce single-stranded DNA  for some reactions (see section seq), the double-stranded nucleic acids had to be separated by heating to 95psy176 C for 3 to 5 minutes. By rapid cooling to 0psy176 C (crashed ice), a renaturation is inhibited. Heat denaturation also removes secondary structures, disturbing a sequencing electrophoresis.
